Title: Resolving the Organ-Dependent Secretome: A Mass Spectrometry Approach to Inter-Organ Communication

Abstract: Exercise produces broad physiological benefits, yet the molecular messengers that coordinate these effects across organs remain poorly defined. To address this gap, we developed a novel mass spectrometry–based framework for high-resolution, quantitative analysis of organ-specific secretomes. This approach enables consistent detection and relative quantification of both soluble and vesicle-associated proteins released directly from individual tissues, allowing us to compare secretion patterns across organs and physiological states. Applying this framework to models of exercise and metabolic remodeling reveals striking tissue-specific signatures and dynamic shifts that point to coordinated inter-organ communication. Distinct changes in the secreted proteomes of muscle, adipose tissue, lung, and placenta highlight the complexity of systemic signaling networks and demonstrate the reproducibility and depth achievable with this workflow. By resolving extracellular proteomes consistently across diverse tissues, this method expands the reach of secretome proteomics and provides a powerful platform for studying inter-organ signaling in vivo.
